Abstract: [Objective] To observe the inhibitory effect of water-soluble substances of stem and leaf of Astragalus membranaceus L. on the central nervous system of mice. [Method] The walking time of the mice, the number of forelimb lifts, and the number of conditioned reflexes in the mice were recorded. [Result] The water-soluble substance of the stem and leaf of Astragalus membranaceus (30g·kg-1 according to the crude drug) can significantly reduce the walking time of the mice, the number of forelimbs, the central excitatory action against sodium benzoate and the strengthening of sodium pentobarbital. Sedative effect with chlorpromazine. [Conclusion] The water-soluble substance of stem and leaf of Astragalus membranaceus has obvious inhibitory effect on the central nervous system of mice, and has synergistic effect with central inhibitory drugs.

Huang Qi was originally contained in Shennong Bencao, a perennial herb of the Labiatae family. Scutellaria Baicalensis Georgi is used as a dry root for medicine, which has the effects of clearing away heat and dampness, purging fire, detoxification, and stopping bleeding. Astragalus root is rich in flavonoids, and modern pharmacological studies have proved that it has many effects such as antibacterial, antipyretic, antihypertensive, hypolipidemic, hepatoprotective and diuretic. Recently, the total flavonoids of stems and leaves of Astragalus membranaceus have also reduced blood fat, blood pressure and anti-arrhythmia. However, the role of stems and leaves of Astragalus membranaceus on the central nervous system has not been reported. In order to provide experimental basis for the development and utilization of stems and leaves of Astragalus membranaceus, the author conducted a preliminary experimental study on the central role of water-soluble substances in stems and leaves of Astragalus membranaceus.
1 Materials and methods
1. 1 material
1.1.1 Experimental animals: Kunming mice, weighing (20 ~ 24g), both male and female, provided by the Experimental Animal Center of Dalian Medical University.
1.1.2 Experimental use: The stems and leaves of Astragalus membranaceus are collected from the 30th Fort in Jinzhou District, Dalian City. They are naturally dried and pulverized. When used, they are decoctioned to make 100% (calculated according to crude drug) solution and placed in the refrigerator for standby; chlorpromazine , Shanghai Hefeng Pharmaceutical Co., Ltd. production, batch number: 020401; sodium benzoate caffeine provided by the Pharmacy Laboratory of Dalian Medical University. Effects on spontaneous activity of mice (walking time and number of lifting of forelimbs): 41 mice were randomly divided into 3 groups, low dose group (10g·kg), high dose group (30g·kg-1), and control group. Give the same volume of normal saline (30 mL / 10 kg). Place the mice in the box during the experiment. Place 1 mouse in a 27 cm × 18 cm × 16 cm box for 5 min before the administration. Record 2 The walking time of the mice and the number of lifting of the forelimbs were normal values. Then, the mice were intragastrically administrated with water-soluble substances of stems and leaves of the scutellaria. After 60 minutes, the spontaneous activity of the mice was measured according to the above method, and the results were expressed by x±s.

Comparisons between groups were made using the medical statistical software (POMS) q test and the χ2 test.

2. The effect of water-soluble substances of stems and leaves of Astragalus membranaceus L. on the spontaneous activity of mice. The results showed that the stems and leaves of Astragalus membranaceus (30g·kg-1) iv made the number of spontaneous activities of mice more obvious than the control group. Reduce, suggest that this product has a central sedative effect.
2.2 Effects of sodium benzoate caffeine (CNB) on exercise excitement Animals were divided into 3 groups, saline group and chlorpromazine group (5 mg·kg - 1 ).
2.3 Effects of subliminal hypnotic dose of pentobarbital sodium The mice were given water-soluble (30g·kg - 1) or NS of the stem and leaf of the scutellaria, and the ith sub-hypnotic dose of pentobarbital sodium 25 mg·kg after 40 minutes. - 1 Observed and recorded the number of cases in which the righting reflex of the mouse disappeared (the disappearance of the righting reflex was maintained for more than 1 min), and the significance test was performed with χ2. The results suggest that the stem and leaf of Astragalus membranaceus (30g·kg-1) can enhance the central inhibition of chlorpromazine.
3 Discussion The root of Astragalus membranaceus is traditionally used, its stems and leaves are discarded, and the research and development and application of ginseng stem and leaf saponins should be further studied to determine its clinical application value. This study found that 30g·kg of water solubles of stems and leaves of Astragalus membranaceus L. significantly reduced the walking time and the number of forelimbs in mice, which was significantly different from the control group (P < 0.01), and the water solubles of Astragalus membranaceus L. 30g·kg- 1 can also significantly inhibit the excitatory state of mice induced by sodium benzoate caffeine, the same dose significantly enhanced the role of subthreshold hypnotic sodium pentobarbital, and significantly enhanced the central inhibition of chlorpromazine. Preliminary results suggest that the water-soluble substance of stem and leaf of Astragalus membranaceus has obvious central sedative effect, and has synergistic effect with hypnotics and antipsychotics. It has been pointed out that Astragalus can reduce the spontaneous activity of mice and enhance the hypnotic effect of sodium pentobarbital on mice. The results of this study indicate that the stems and leaves of Astragalus membranaceus also have a central inhibitory effect similar to their roots. Modern pharmacological studies have found that stems and leaves of Astragalus membranaceus are rich in sutenasin, which is as high as 3% to 10%. It can be considered that the stems and leaves of Astragalus membranaceus have obvious biological activity, and it is worthy to conduct in-depth research on the flavonoids they contain for development and utilization.
